For me the highlights are:
- moving newsletters and mass mailings into a separate folder. This keeps my inbox clean.
- the ability to easily block emailers that you never want to hear from again (the blackhole)
- the ability to send reminders in the future. When responding to someone, I often put a date (e.g., oct23@sanebox.com) in the bcc field and the email comes back to me at that date. This way I can ensure follow-up. 

There are so many other features, but those are the ones I use daily.
